I need some type of padding to make a holster for my purse. What is the best for that?

Fusible Fleece would be perfect for that, this is high loft so it will give volume and stability to your piece.

Would this work for the inside of a soft eyeglass case?

Yes, That is what I make fabric eye glass cases and small fabric wallets.

Would this work to put inside footie jammies to cover holes

It is very thin but might work temporarily.

I'm making placemats. Is this product thin enough that they would lay flat?

It would depend on how you are finishing the edges. Seams will obviously be higher than the body of your work, but bias edging lays flat very well.
