I am an older female. Can I set this up by myself?

Yes, you should be able to. I am a 75 year old female myself and like the tent because I can manage it by myself. If you have balance issues, you should be careful, because it flips open with a bit of force, and then you have to pop out the sides and lift it into place. Not heavy, just a little awkward.

How to use guide lines. It’s blowing in

I haven't tried the guide lines. You can just pop the sides back in place, but it's true that if there is much wind it's not very stable. The good side of that is how easy it is to collapse and take inside. Sorry, I guess I'm not much help in answering your question.
