It does not work on plastic. The product is activated by metal ions, so it fully cures only in presence of metal.

will this work for diesel exhaust manifold block off plate with high temps?

This product can only resist up to 300-350 degrees F, not recommended for exhaust manifolds. The metallic gasket is the only thing that will create a proper seal for those temps of a diesel engine.

Can I use 515 to seal around the leak for automatic transmission fluid around the dipstick tube as it connects to the chevy automatic transmission?

515 is primarily an anaerobic, used to prevent air leakage between torqued surfaces. I have used it on gear cases and power equipment transmissions with success. It may work on your dipstick, if you can secure it so it doesn't move around. Of all things JB weld may be a better repair.
