How long does it take to charge by USB?

It dosen't charge. The USB port is the only port that the lamp have, so is there where you must plug it into the wall socket, with the cord, that comes with it.David

Can this replace a desk lamp?

Not a desk lamp for writing. Its pretty muted with the yellow and the cutouts that are brighter point up to project on the wall. I'd put it more in a nightlight range...purely decoration.

How long does the light last before it goes out

I am not entirely sure what you are asking. Are you asking if the light turns off on its own or how long it takes for the bulb to burn out? The light bulb does not turn off on its own. You have to manually turn it on and off each time the light is in use. As far as the life of the light bulb, I am unsure. I can tell you this, we bought the light in mid September and have used it every night since for at least 8 hours and it still lights up. The light doesn't seem to have dulled over time either. I hope this helps.
