Yes they do work for the Ruger gp100 7 shot. They are a perfect fit. Just ordered 2 more myself.

How does this work with a taurus 66?

As long as you are using .38 or 357 magnum bullets and has 7 shots, I think you're good. In fact, taurus 66 is also in the list.

Will this work with 357 Smith and Wesson revolver 6 shot?

You'll need to purchase the HKS 586-A for your S&W 686, 6 shot. The 587-A is for a seven round 686.
