Will it work on 13" tire

Yes, but I had a two piece rim. Works fine holds air.

Will this tube work in a rim that has stem hole offset from center?

Most wheelbarrows and other things that use these type wheels are generally offset to the side of the rim. Once you put the valve through the hole you'll need to hold it there while mounting the tire back over it. Half the tire should be on the rim and the tube inside the tire. Then hold the valve in place as you put the rest of the tire onto the rim. HTH
